Abstract

The present attempt was focused on the molecular characterization of certain wild medicinal plants growing in and around Banasthali region. In this context five medicinal plant species (viz. Emblica officinalis, Butea monosperma, Citrus limon, Dalbergia sissoo, Eucalyptus globulus) were selected from the natural flora of Banasthali region. These plants were subjected for preparation of DNA fingerprint profile for authentic documentation and characterization of germ plasm for their conservation. The total genomic DNA of selected plant species was extracted using the eTAB method with some modification. This consistently achieved high quality; low polysaccharide type DNA in sufficient amount from fresh leaf tissue of selected plant species and enabled us to undertake further work on molecular profiling. Based on the specificity of genotype of a plant, a particular DNA profile can be ascribed to a particular plant. The wild medicinal plants of Banasthali region were characterized at molecular level with nine random primers. Among them the primer-5 seems to be more informative as it generated maximum average number of bands in selected plants. The present work has contributed in the characterization of wild medicinal plants and provides valuable information on authentication of medicinal plants of this region through their RAPD pattern.
