Effect of dietary supplementation of different selenium sources on semen quality and oxidative stress in frozen-thawed semen of bucks

Abstract

Twenty-four Barbari bucks with similar semen quality were selected and divided into four equal groups. Bucks in control were maintained without selenium (Se) supplementation and in treatment supplemented different Se sources @ 0.30 mg/kg viz., inorganic Se as sodium selenite (ISe), organic Se as Se-enriched yeast (OSe) and nano selenium (NSe) for a period of 90 days. Two ejaculates per week per buck were collected from day 91 to 104. The collected semen was cryopreserved and evaluated 7 days after post freezing. The sperm viability (%) was significantly (P<0.05) higher in NSe (67.60) compared to ISe (60.00) and OSe (61.80). Sperm with intact acrosome (%), hypo-osmotic swelling test positive (%) and sperm with normal membrane fluidity (%) were significantly (P<0.05) higher in NSe (56.20, 68.23 and 55.50) than ISe (46.80, 56.79 and 47.80) and Ose (48.70, 58.67 and 48.20). Sperm mitochondrial activity (%) was significantly (P<0.05) higher in NSe (61.40) compared to ISe (54.10) and OSe (55.20). Total motile sperm (%) and progressive motile sperm (%) were significantly higher in NSe (60.14 and 41.31) than ISe (51.57 and 36.14) and OSe (53.76 and 37.12). Lower values (P<0.05) of antioxidant enzymes viz. glutathione peroxidase (units/ml) and superoxide dismutase (units/mg protein) were recorded in NSe (3.709 and 13.219) than ISe (4.604 and 14.749) and OSe (4.305 and 14.289). Hence, nano form of Se can be recommended for dietary supplementation of breeding Barbari bucks for better post-thaw semen quality.
