Tissue Culture Studies for Micropropagation of Pelargonium graveolens L’ Herit

Abstract

An efficient protocol has been developed for direct regeneration of Pelargonium graveolens L’ Herit. Murashige and Skoog (MS)basal medium supplemented with 2.0 mg l–1 BAP and 1.0 mg l–1 IAA was optimum for attaining maximum number of shoots (12±0.94) having 60% regeneration. In vitro grown shoots of direct origin were transferred to ½ strength MS medium fortified with 1.0 mg l–1 IBA for attaining the maximum number of roots (11.0 ± 1.65) having 60% regeneration. Regenerated plantlets with well-developed shoots and roots were successfully transferred to soil. The study demonstrated an efficient in vitro procedure for micropropagation via nodal explants in P. graveolens L’ Herit which could be useful for large scale proliferation of this cash crop. Essential oil of graveolens L’ Herit is widely used in high grade perfumery, cosmetic industries, as a flavouring agent in many food categories and soft drinks. The oil has anti-bacterial and insecticidal properties and is profusely used in aroma therapy.
