α-Glucosidase inhibitory activity of styrylpyrone and flavonoids isolated from Alpinia mutica Roxb. seed

Abstract

The seven major phytochemicals, farnesol (1), 5,6-dehydrokawain (2), pinocembrin (3), cardamomin (4), naringenin (5), pinocembrin chalcone (6) and alpinetin (7) were isolated from seed extract of Alpinia mutica. The structures were characterized using NMR spectroscopic data and HRESIMS. These isolated compounds were tested for their inhibition of α-glucosidase activity. Compound 5 was found with most potent inhibitory activity with IC50 value of 8.77 ± 1.04 μM and compound 3 also showed high activity with IC50 value of 62.77 ± 2.18 μM. Furthermore, the study on their biosynthetic origin of isolated compounds revealed that styrylpyrone and flavonoids from A. mutica seed originate from cinnamoyl CoA or p-coumaroyl-CoA with malonyl-CoA chain extension. Moreover, compound 6 was the precursor for flavonoids formation of A. mutica seed and this is the first report on the isolation of importance precursor, 6 from A. mutica.
