Phytochemical analysis and anticancer activity of leaf, bark, and seed extracts of Gymnacranthera farquhariana (Hook. f. & Thomson) Warb.

Abstract

The members of family Myristicaceae are widely used group in the ethnomedical system. However, no prominent studies have been reported on Gymnacranthera farquhariana. The current study aims to examine the phytochemical composition and cytotoxic activity of G. farquhariana leaf, bark, and seed in aqueous and methanolic solvents. These extracts were screened for the presence of phytochemicals and tested for cytotoxic and anticancerous activity with HEK293 and MCF 7 cell lines. The methanol seed extract was subjected to LCMS analysis. The result revealed the presence of carbohydrates, proteins, alkaloids, cardiac glycosides, flavoanoids, saponins, tannins, anthraquinones, glycosides, reducing sugars, phenolics, and phlobatanins in methanol and aqueous extracts. The protein content was maximum in the bark extract of aqueous solvent (8.524±0.85 mg/g). Phenolic content was higher in methanolic seed extract (1.53±0.024 mg/g), reducing sugars in methanolic leaf extract (5.346±0.391 mg/g), and tannin also in methanolic seed extracts (2.075±0.016 mg/g). The seed extract in methanol has exhibited higher anticancerous activity than leaf and bark against MCF7 cell line (1.86±0.076 at 1000 μg/mL) and less cytotoxicity on HEK293 (30.47±4.39 at 1000 μg/mL) in aqueous leaf extract. The LCMS profiling has led to identification of 10 anti-breast cancer compounds in methanol seed extract. The present study has indicated the usefulness of G. farquhariana for developing therapeutic compounds for anticancerous purposes.
