Online published on 29 June, 2023. Abstract Silver nanoparticles are promising candidates for use as anti-inflammatory, antimicrobial, antiviral, or antitumoral potential. The present work was designed to investigate the potential of silver nano formulation of Tabernaemontana divericata for anti-inflammatory potential. Solubility and bioavailability of phytoconstituents is the major problems, which can be overcome with nano formulation. Nanosizing of phytoconstituents will help to increase the solubility ultimately increase the pharmacological potential. Cotton pellet granuloma, carrageenan-induced and formalin induced paw edema were used to assess the anti-inflammatory potential of T. divericata nano formulation. Results clearly demonstrated that nano formulation can be used as a potential source for natural anti-inflammatory drugs. T. divericata nano emulsion (Td-Ne) was evaluated for particle size, PDI, zeta potential, SEM, XRD, DSC and for its anti-inflammatory potential with cotton pellet granuloma, carrageenan-induced and formalin induced paw edema in rats. Average particle of Td-Ne was found to be 143.33 nm, PDI 0.764 nm, zeta potential -97.4 mV and XRD showed the characteristic peaks at 29 θ, 27.88 θ and 27.12 θ. SEM of Td-Ne showed spherical shape and smooth surface of nanoparticles present in formulation, and melting endotherm of Td-Ne was observed in DSC at 333.8°C. Td-Ne (50 and 100 mg/kg bd. wt) exhibited significant anti-inflammatory action with cotton pellet granuloma (p<0.0001, p<0.05), carrageenan induced (p<0.0001, p<0.05) and formalin induced inflammation (p<0.0001, p<0.05). T. divericata nano emulsion showed significant anti-inflammatory potential; can be utilized for managing the inflammatory diseases with improved solubility and bioavailability.
