Online published on 24 August, 2018. Abstract Diseases caused by Salmonella enterica serovars are particularly prevalent in developing countries. It is considered one of the major pathogens of public health significance worldwide. Genotypic characterization based on the analysis of restriction fragment length polymorphism of the fliC gene (fliC PCR-RFLP), was performed on 31 Salmonella isolates from poultry which includes Salmonella enteritidis (51.6%), Salmonella typhimurium (25.8%), Salmonella infantis (19.4%) and Salmonella colindale (3.2%). DNA of 31 Salmonella strains was extracted using phenol-chloroform-isoamylalcohol (25: 24: 1) method. Extracted DNA was used as template for amplifying fliC gene (1500bp) using specific primers. PCR products were subjected to digestion using Sau3AI restriction endonuclease. All digested DNA were electrophoresed on agarose gel. PCR-RFLP results showed 3 patterns. Our results showed that PCR-RFLP technique with Sau3AI restriction endonuclease was able to differentiate Salmonella enteritidis and Salmonella typhimurium.
