Characterization of colistin resistance gene MCR-1 in Klebsiella Pneumoniae

Abstract

Colistin is used to treat infections with multidrug-resistant bacteria such as Klebsiella pneumoniae. However increased use of Colistin resulted into resistance in some of the bacterial strains to it. Present investigation was undertaken to find out the prevalence of Colistin-resistant gene (mcr-1) in multidrug resistant (MDR) Klebsiella pneumonia, collected from clinical samples of different Critical care units (CCUs).The study was conducted in the Department of Microbiology and Medicine at Rama Medical College Hospital and Research Centre (RMCH&RC), Kanpur for 18 months. (March 2022 to September 2023). Klebsiella pneumoniae species were examined for Colistin susceptibility by the Broth Micro dilution (BMD) method, and the minimum inhibitory concentration (MIC) was determined. The drug resistant gene (mcr-1) was detected by adopting conventional PCR. Out of 240 Klebsiella pneumoniae isolates, 99 (41.3%) isolates were found to be Colistin resistant, which were obtained from ICU. Prolonged hospital stay and direct antibiotic pressure in the hospital might have led to the development of carbapenem-resistant Klebsiella pneumonia (CRKp) variants.
