Sub-chronic Indoxacarb-induced haemato-biochemical and histo- pathological changes in rats

Abstract

Indoxacarb is a broad spectrum oxadiazine insecticide with selective action on insects. The present study was conducted to assess the indoxacarb-induced sub-chronic toxicity in rats for 60 days. Male Wistar Rats were divided into four groups of six animals each. Control group received corn oil orally at 0.5 ml/kg body weight. Rats in three treatment groups received indoxacarb mixed in corn oil orally at 15 mg/kg (1/40th MTD), 30 mg/kg (1/20th MTD) and 60 mg/kg (1/10th MTD). Indoxacarb at different dose levels caused significant increase (P<0.05) in alanine aminotransferase (ALT) and blood urea nitrogen (BUN) levels compared to control group. Aspartate aminotransferase (AST), serum creatinine and serum cholesterol levels were increased significantly (P<0.05) only in medium and high dose groups. Total erythrocyte count (TEC), packed cell volume (PCV) and hemoglobin (Hb) showed significant (P<0.05) decrease in 60mg/kg indoxacarb group compared to control and other groups. Dose dependent significant (P<0.05) increase in total leucocyte count (TLC) was observed in all the indoxacarb treated groups compared to control. Histopathologically, liver of 60 mg/kg group showed severe degenerative and fatty changes of hepatocytes after 60 days of exposure. Spleen showed severe lymphoid hyperplasia of white pulp in all the indoxacarb treated groups. In brain, both cerebrum and cerebellum showed mild to moderate neuronal degeneration with severely engorged meningeal blood vessels and brain capillaries in 60 mg/kg test group. Occasionally, satellitosis in cerebrum and Purkinje cell degeneration in cerebellum were observed. In conclusion, sub-chronic exposure to indoxacarb causes severe systemic toxicity in rats.
