Pathological changes in doxorubicin induced toxicity and its ame-lioration in Wistar rats

Abstract

The present study was carried outin 24 female rats divided in 4 groups: Group 1-Control; Group 2-doxorubicin-Toxic control @ 2mg/kg body wt. by intravenous injection for 5 days followed by weekly once for 2 weeks; Group 3-Pretreatment with ascorbic acid @ 500mg/g feed by enteral route for 7 days followed by intravenous injection of doxorubicin like in group 2, and continued ascorbic acid supplementation for 19 days; Group 4-Pretreatment with Spirulina @ 1000mg/kg feed by enteral route for 7 days followed by intravenous injection of doxorubicin like in group 2, and continued Spirulina supplementation for 19 days of experimentation. Histopathologically, heart from group 2 exhibited degeneration, necrosis and haemorrhages and moderate focal infiltration of lymphocytes. Kidneys revealed severe interstitial congestion, haemorrhages and marked degenerative changes of tubular epithelium with epithelial casts in the lumen. Liver showed congested sinusoids and central veins, fatty changes and focal necrosis of hepatocytes andbile duct hyperplasia. The ameliorative groups 3 and 4 showed mild to moderate improvement. Tissue enzyme as says revealed a significant (P<0.05) increase in thiobarbituric acid reactive substances (TBARS) and significant (P<0.05) decrease of reduced glutathione (GSH) and superoxide dismutase (SOD) activities in group 2. The ameliorative groups 3 and 4 exhibited mild to moderate improvement in all parameters compared to group 2. Results showed that vitamin C @500mg/kg feed was less protective and Spirulina @1000mg/kg feed offered moderate protection in counteracting the toxic effects of doxorubicin.
