Abstract This study investigates the lipid-lowering and hepatoprotective potential of hydroethanolic extract of Cichorium intybus (HECL) against lead and nickel-induced lipid profile alterations and hepatic injury in rat model. The effects of lead and nickel exposure on serum lipid profiles and liver enzymes were analyzed, and the protective role of HECL was examined. Exposure to lead, nickel, and their combination led to significant (p<0.05) increases in triglycerides (TRI), total cholesterol, and low-density lipoproteins (LDL), while high-density lipoprotein (HDL) levels were significantly reduced. These changes were indicative of disrupted lipid metabolism and were consistent with previous findings. Elevated activities of liver enzymes (ALT, AST, ALP, GGT, and LDH) further underscored hepatocellular damage due to metal toxicity. HECL administration demonstrated a significant (p<0.05) reduction in elevated enzyme levels induced by lead and nickel exposure, thereby exhibiting hepatoprotective effects. The extract's ability to enhance lipid profiles and elevate HDL levels reinforces its potential as a lipid-lowering agent. Mechanistically, HECL's polyphenolic flavonoids and antioxidant properties contribute to membrane stabilization, counteracting oxidative stress and promoting liver health. This research substantiates the multifaceted benefits of Cichorium intybus extract, offering insights into its potential to counteract heavy metal-induced lipid profile disturbances, and hepatocellular damage. The study underscores the importance of natural interventions in mitigating the adverse effects of environmental toxins and points towards HECL's promising role in enhancing metabolic and cardiovascular well-being.
