PK/PD Integration and pharmacokinetic study of moxifloxacin after single intravenous and intramuscular administration in female sahiwal calves

Abstract

Moxifloxacin is a fourth generation fluoroquinolone discovered in 1999, with activity against a broad spectrum of pathogens, encompassing Gram-negative and Gram-positive bacteria. The present research was aimed to study the PK/PD integration and pharmacokinetic profile of moxifloxacin at the dose of 5 mg.kg-1 body weight in calves following single intravenous and intramuscular administration in female sahiwal calves. Moxifloxacin concentration in plasma samples were determined by microbiological assay using Escherichia coli MTCC 443. The plasma concentration- time profile following intravenous and intramuscular administration in calves was best described by two-compartment open model and one-compartment open model, respectively. Following intravenous administration the plasma drug concentration of 11.70 ± 0.21 μg.ml-1 was observed at 0.04 h and 0.21 ± 0.00 μg.ml-1 at 36 h. Cmax:MIC ratio was found 47.20 and AUC: MIC ratio was found 186.63. After intramuscular administration of moxifloxacin, the drug concentration of 0.35 ± 0.07 μg.ml-1 was observed at 0.04 h which gradually increased and reached at peak plasma concentration (Cmax) of 3.05 ± 0.25 μg.ml-1 at 1.70 ± 0.12 h (tmax) and Cmax:MIC ratio was reported 12.20 while AUC:MIC ratio 173.22. Integration of pharmacokinetic data generated from the present study and minimum inhibitory concentration (MIC) suggest that moxifloxacin can be administered at the dose of 5 mg.kg-1 body weight through either intravenous or intramuscular route at the dose interval of 24 h to combat susceptible bacterial infections in calves.
